OCT Compound 118ml
Embedding medium for frozen tissue specimens.
OCT Compound is a water-soluble blend of glycols and resins that provides a convenient specimen matrix for cryostat sectioning at temperatures of -10?C and below.
OCT leaves no residue on slides during staining procedure, eliminating undesirable background staining.
Dissected tissue, mounted in OCT Compound can be frozen between -20 and -80?C.
Developed and optimized for chromogenic IHC experiments, OCT Compound will not dull microtome knives.
OCT Compound contains:
- Polyvinyl alcohol
- Polyethylene glycol
- Nonreactive ingredients

Epoxy resins and methacrylates are often harmful. Resin blocks are harmless after curing but, if cured blocks are sawn, make sure that the dust is not inhaled.
Agar 100 – replaces Epon 812 resin, now unobtainable. The formulation of Agar 100 resin is the same as Epon, but is less viscous than many samples of Epon 812 and is more closely controlled in its manufacture. Epoxide equivalent between 148 and 150.
ERL 4221 has replaced ERL4206. It has a higher viscosity (180cps) than the previously used ERL 4206 (60cps). Ref: Ellis, E Ann, Solutions to the Problem of Substitution of ERL 4221 for Vinylcyclohexene dioxide in Spurr Low Viscosity Embedding Formulations, Microscopy Today, Vol 14, No 4, July 2006.
Technical Note
The original formulation, ERL 4206 (VCD) – DER® 736 embedding medium devised by Spurr (1969) combined very low viscosity components for rapid specimen penetration.
This embedding medium has been especially suitable for a variety of mineral and biological specimens. Since ERL 4206 is no longer available, ERL 4221 has been substituted as a replacement*.
It will work well for most users but it does have somewhat higher viscosity (180cp vs. 60cp) than the ERL 4206, which can impact some protocols employing vacuum embedding.

A block of MEDIUM hardness can be obtained by using the following formula based on a batch of 10 grams without the flexibilizer, DER 736:

A harder block can be produced by decreasing the DER® 736 component to 0.95g in the above formula; a softer block by increasing the DER 736 to 1.90g.
ALL STEPS MUST BE PERFORMED UNDER A HOOD AND GLOVES MUST BE WORN FOR PROTECTION.
Preparation:
Care should be taken to weigh the ingredients accurately into a beaker. The quantities given in the first column are for firm blocks.
For harder blocks, decrease the DER 736 to 0.95g; for softer blocks, increase the DER 736 to 1.90g.
For rapid cure, increase the DMAE. It is recommended to add the catalyst (dimethylaminoethanol) last, after having carefully mixed the other components.
If bubbles are a problem, they may be eliminated by placing the beaker into a desiccator under a gentle vacuum.

Quetol 651 500g
This is an epoxy resin with a viscosity of 15 cps at 25 C and an epoxy equivalent of 115. It is miscible with water, alcohol and acetone.
When used with NSA and DMP-30, it forms a low viscosity mix which penetrates tissue easily.

Accelerators for epoxy resins
BDMA is recommended in preference to DMP-30 since it is less viscous and has a longer shelf life.
Accelerators should be stored dry in a desiccator, but need not be in a refrigerator.
All these materials have a shorter shelf life than the resins and should be discarded after six months.
DMP-30 is harmful
BDMA, S-1 + accelerator (LV) are corrosive.

A variety of film-forming solutions are available to prepare support films and replicas of smooth surfaces.
Formvar (polyvinyl formal) is normally used as a 0.25% solution in chloroform.
The thickness and stability of the film can be altered by adjusting the concentration between 0.25% and 0.5%.
Pioloform is polyvinyl butyral dissolved in chloroform (0.3 – 1.5%).
The solution should be stored in a refrigerator in an amber glass bottle, and brought up to room temperature before use.
Butvar B98 is a polyvinyl butyral resin containing 20% polyvinyl alcohol, and is normally used as a 0.25% solution in chloroform.
It is an alternative support film to Formvar as reported by: Handley and Olsen, Ultramicroscopy 4, p479 – 480, 1979.
Films made from Butvar are hydrophilic, and are therefore suitable for negative staining methods, having good mechanical stability, high electron transparency and minimum intrinsic structure.
Butvar B98 can also be applied to grids as an adhesive.

Hexamethyldisilazane (HMDS) 100ml
HMDS, [(CH3)3Si]2NH, can be used in place of critical point drying for the preparation of soft tissue for SEM examination, for example, of delicate insect tissues.
It is faster, preserves surface detail, reduces thermal and pressure stresses, and may also reduce the extraction of cellular components compared with CPD. HMDS can be used to dry specimens such as bacteria on polycarbonate filters. 100ml

Paraplast Plus 1kg
Paraplast Plus is a double filtered paraffin embedding medium contains plastic polymers of regulated molecular weights and a small percentage (0.8 %) of dimethyl sulphoxide (DMSO) to penetrate tissues three times faster than standard media.
It melts rapidly at 56 °C, and allows short ribbons of sections cut to 1 m, and excellent ribbon continuity with 2 m thicknesses.
